Tips Menjawab Reviewer Jurnal Internasional, Contoh “Response to Reviewers”

Reviewer #1
The author’s has explained well the extraction process of the pectin from the mangosteen rind and examined the morphological and pharmacological properties of the pectin. However, there are things that need to be corrected to be accepted.
Replies to Reviewer #1
First of all, we deeply appreciate your helpful comments. Our replies to the reviewer’s inquiries and revised points are as follows. Please confirm the relevant parts highlighted in red in the revised manuscript.
Comment 1:
The author must describe when the rind sample is taken (which development stage of fruit), maybe during fruit development will be different in the amount of pectin.
Reply to comment 1:
We have revised the materials section. This was stated on page 4 lines 4. The mangosteen fruit was harvested after 100 days of the flowers blooming.
Comment 2: Phenolphthalein and phenol are not the same, but they are very similar. the author must correct the use of the term.
Reply to comment 2:
Thank you for your valuable comment. We used only phenolphthalein. We have revised and stated on page 5, lines 3 and 7 from the bottom.

This work investigated the characterization of the pectin from Indonesian Mangosteen, followed by its antioxidant activity invitro. It is a routine work from local resouces. However, some concerns should be considered.
Replies to Reviewer #2
First of all, we deeply appreciate your helpful comments. Our replies to the reviewer’s inquiries and revised points are as follows. Please confirm the relevant parts highlighted in red.
Comment 1: Is there a national standard of pectin in Indonesia? Does the pectin here meet the standard? It is very important. The characterization should follow the standard for application.
Reply to comment 1: Yes, there is a national standard of pectin in Indonesia namely Indonesian Pharmacopeia. However, it was stated only methoxyl level and galacturonic level which was same as Handbook of Pharmaceutical Excipients. For other standards, we used previously paper published. As reviewer suggested, we have revised the results section. This was stated on page 8, lines 5-7 from the bottom.
Comment 2: The submission is just a list of results about a new pectin. It would be better to concern the internal connection between different parts.
Reply to comment 2: Thank you for your valuable comment. We have added sentences to concern the internal connection between different parts. This was stated on the results and the discussion sections highlighted with red color (page 6, lines 5,7 from the bottom, page 7, lines 5,12-13 from the bottom, page 8, lines 6, page 9, lines 1-2.

The manuscript reported by Dr. Wathoni represented the effect of sacran hydrogel film containing keratinocyte growth factor against wound healing in vtiro and in vivo. Most of the study is well designed and the scientific value is given enough. However, the authors did not show the literature the authors have recently reported (Wathoni N, Motoyama K, Higashi T, et al. Enhancement of curcumin wound healing ability by complexation with 2-hydroxypropyl–cyclodextrin in sacran hydrogel film. Int J Biol
Macromol (Netherlands), May 2017, 98 p268-276). This paper is strongly related with present work. To make clear the novelty of this paper, the authors have to state the differences between the present work and previous work. In addition, although this is my thought, you don’t have to show any appendix written by Japanese (PPT file).
Furthermore, it would be helpful to determine the effect of KGF concentrations in the hydrogel film against wound healing in vivo.

Reply to comment 1:
In our previous study, Sac-HF can be used as an effective wound dressing biomaterial to deliver curcumin through the complexation with HP–CyD. These results suggest that
a soluble drug in Sac-HGF has a potential as a new transdermal. Therefore, we have revised the introduction. This was stated on page 4, lines 13-16.
Comment 2: You don’t have to show any appendix written by Japanese (PPT file)
Reply to comment 2:
As the reviewer suggested, we have removed Japanese appendix.
Comment 3: It would be helpful to determine the effect of KGF concentrations in the hydrogel film against wound healing in vivo.
Reply to comment 3:
Regarding the effect of KGF concentrations in the hydrogel film against wound healing in vivo, in this study the dose (100 ng) was fixed based on the previous study
[Sotozono C, et al. Paracrine role of keratinocyte growth factor in rabbit corneal epithelial cell growth. Exp Eye Res. 1994 Oct;59(4):385-91 PMID: 7859813]. In addition, the present study focused on the potential delivery of KGF in Sac-HF as wound healing therapy. This was stated on page 5, lines 15.
Minor comments
Comment 1: Fig 1B: Please add more explanation about each spectrum. And fibroblast growth factor is a protein, right? This peak seems to melting point, not degradation? Please clarify it again.
Reply to comment 1: As the reviewer suggested, Fig. 1B has been revised. In addition, fibroblast growth factor is a protein, and the peak is melting point as mentioned in page 8.
Comment 2: 3.2 line 8 from the bottom: typo “a¬”
Reply to comment 2: Word “a” has been deleted.
Comment 3: 3.5. line 11: typo “plays¬”
Reply to comment 3: The word “plays” had been changed to “play”. This was stated on page 11, line 11.
Comment 4: 3.6. line 6: typo “stimulates¬”
Reply to comment 4: The word “stimulates” had been changed to “stimulate”. This was stated on page 11, line 9.

The authors prepared sacran (water-soluble polymer) hydrogel-based film for the treatment of wound healing. The sacran film contains KGF, and the inclusion of the growth factor showed the wound healing by using in vitro and in vivo mouse model. I think that the film has useful points for the treatment of wound healing. I have several comments about the manuscript.

First of all, we deeply appreciate your helpful comments. Our replies to the reviewer’s inquiries and revised points are as follows. Please confirm the relevant parts highlighted in red.
Major points
Comment 1: Sacran-based hydrogel film may have good property in the point of film. However, I do not understand the usefulness of the compound compared to typical water-soluble polymers such as HPMC, HPC and/or PVA. These polymers are well-known medical excipients. So, the discussion with the polymers is necessary to show
the novel point.
Reply to comment 1: Thank you for your valuable comment. As the reviewer suggested, we have revised introduction as follows: Nowadays, a hydrogel film for wound dressing application has gained a lot of attention due to their excellent properties like impermeability to bacteria, film transparency, and less painless 23).
Many hydrogel films are constructed by synthetic or semi-synthetic polymers, such as hydroxy propyl methyl cellulose, polyurethanes and/or polyvinyl alcohol 25,27,29).
Natural polysaccharides have been known as biopolymers for wound dressing materials which have biomedical activities and a lower toxicity than synthetic or semi-
synthetic polymers 24). This was stated on page 4, lines 3-8.
Comment 2: Did you check the cytotoxicity about KGF-loaded sacran film in vitro?
Reply to comment 2: As the reviewer pointed out, we have newly performed a cytotoxicity study. We have added the method and the result of the cell proliferation
study in 2.7. Proliferation Study on page 6 and Fig. 3, respectively. In addition, we have added the statements on page 9, lines 18-24.
Minor points
Comment 1: Can you add the typical culture condition of cell culture and the
information of cells and reagents with the section of “cell culture”?
Reply to comment 1: 1. As the reviewer’s suggestion, we have added the information in cell culture section. Those was stated on page 5, 2.2.
Comment 2: According to 2.3., how did you add or apply KGF into the film? What kind of aqueous solution was used to disperse KGF?
Reply to comment 2: As the reviewer pointed out, we have added the following statements: KGF solution (50 ppm KGF suspended in ammonium sulfate solution) and 10 L of KGF, respectively. These were stated on page 4, line 3 from the bottom and page 5, line 6 in the 2.4 Preparation of Sac/KGF-HF section.
Comment 3: Japanese supplementary information should be removed.
Reply to comment 3: According to the reviewer’s suggestion, we have removed it.
